CEBPA (CCAAT enhancer binding protein alpha)
نویسنده
چکیده
C/EBPa protein domains The basic region leucine zipper domain mediates DNA binding, homodimerization of C/EBPa and heterodimerization with other members of the C/EBP family. This region is also involved in mediating protein-protein interactions with other transcription factors involved in lineage determination and growth proliferation. N-terminal region transactivating domains mediate interactions with transcriptional machinery and proteins important in cell cycle control. The CEBPA mRNA gives rise to two different translational isoforms by using different start codons within the same open reading frame by means of leaky ribosome scanning; the full length 42 kDa protein and a 30 kDa truncated form. These isoforms display contrasting functions in regards to gene activation and cell proliferation. Both isoforms can be detected within the cell and it is likely that the ratio of isoforms is important in mediating proliferation and differentiation control.

Molecular Pathways Complexity of CEBPA Dysregulation in Human Acute Myeloid Leukemia
The transcription factor CCAAT enhancer binding protein alpha (CEBPA) is crucial for normal development of granulocytes. Various mechanisms have been identified how CEBPA function is dysregulated in patients with acute myeloid leukemia (AML). In particular, dominant-negative mutations located either at the Nor the C terminus of the CEBPA gene are observed in roughly 10% of AML patients, either ...
متن کاملCEBPA-regulated lncRNAs, new players in the study of acute myeloid leukemia
CCAAT/enhancer-binding protein-α (CEBPA) is a critical regulator of myeloid differentiation. Disruption of CEBPA function contributes to the development of acute myeloid leukemia (AML). CEBPA regulates a large number of protein coding genes of which several were shown to contribute to CEBPA function. In this study, we expand the analysis of CEBPA transcriptional targets to the newly identified ...
متن کاملIdentification of a novel distal enhancer in human adiponectin gene.
Adiponectin is exclusively expressed in adipose tissue and secreted from adipocytes, and shows anti-diabetic and anti-atherogenic properties. However, the precise transcriptional mechanism of adiponectin remains elusive. In this study, the 5' flanking promoter region of human adiponectin gene was analyzed using UCSC genome browser, and a 10 390-bp fragment, containing an evolutionally conserved...
متن کاملDeficient CEBPA DNA binding function in normal karyotype AML patients is associated with favorable prognosis.
CCAAT/enhancer binding protein-α (CEBPA) mutations in acute myeloid leukemia (AML) patients with a normal karyotype (NK) confer favorable prognosis, whereas NK-AML patients per se are of intermediate risk. This suggests that blocked CEBPA function characterizes NK-AML with favorable outcome. We determined the prognostic significance of CEBPA DNA binding function by enzyme-linked immunosorbent a...
متن کاملC/EBPα regulated microRNA-34a targets E2F3 during granulopoiesis and is down-regulated in AML with CEBPA mutations.
The transcription factor, CCAAT enhancer binding protein alpha (C/EBPα), is crucial for granulopoiesis and is deregulated by various mechanisms in acute myeloid leukemia (AML). Mutations in the CEBPA gene are reported in 10% of human patients with AML. Even though the C/EBPα mutants are known to display distinct biologic function during leukemogenesis, the molecular basis for this subtype of AM...
متن کاملThe CEBPA gene is down-regulated in acute promyelocytic leukemia and its upstream promoter, but not the core promoter, is highly methylated.
Impairment of CCAAT Enhancer Binding Protein alpha (CEBPA) function is a common finding in acute myeloid leukemia; nevertheless, its relevance for acute promyelocytic leukemia pathogenesis is unclear. We analyzed the expression and assessed the methylation status of the core and upstream promoters of CEBPA in acute promyelocytic leukemia at diagnosis. Patients with acute promyelocytic leukemia ...
